Abstract

Diabetes Mellitus increases free radical rate and reduces oxidant defense system. One of patho-mechanisms due to increased free radical and reduced oxidant defense system relates to the increase of lipid peroxidation process and the decrease of erythrocyte membrane tenacity. Kernel Guava (Psidium guajava L) has antioxidant potential because it contains -carotene, -tocopherol (vitamin E) and higher vitamin C. Therefore, kernel guava neutralizes free radical, as expected to inhibit lipid peroxidation and to improve erythrocyte membrane tenacity in Diabetes Mellitus. Research Objective to confirm that Kernel Guava (Psidium guajava L) juice inhibits lipid peroxidation and increases erythrocyte membrane tenacity at Diabetes Mellitus treated Rat. Research considers 25 male rats of Rattus novergicus Strain Wistar type, with initial weight of 100-200 grams and 2-3 months of age. The rats divide into five groups: (1) Normal group; (2) Diabetes Mellitus group; (3) Diabetes Mellitus group with kernel guava juice therapy dosed for 0.5 g/rat/day; (4) Diabetes Mellitus group with kernel guava juice therapy dosed for 1 g/rat/day; (5) Diabetes Mellitus group with kernel guava juice therapy dosed for 2 g/rat/day. At day 10, rats have been sacrificed for analysis. Lipid peroxidation process at erythrocyte membrane has been acknowledged through MDA rate (mol/ml) measured by Thiobarbituric Acid (TCA) test. Erythrocyte membrane tenacity evolves from the measurement of hemolytic rate (%) and abnormal erythrocyte rate (%). Results of research need statistic test, including ANOVA, regression, and correlation, all with SPSS. Results of research : all doses of kernel guava juice therapy (0.5;1;2 gr/rat/day) reduce MDA rate (mol/ml) on average (2.56  1.08), hemolytic rate (%) on average (67.49  2.61), and abnormal erythrocyte rate (%) on average (51.79  2.87), when Normal group seems compared to Diabetes Mellitus group. Results of statistic analysis indicate that kernel guava juice therapy with all doses (0.5;1;2 gr/rat/day) significantly inhibits lipid peroxidation and improves erythrocyte membrane tenacity of Diabetes Mellitus treated rat. Results of research determine that Kernel Guava (Psidium guajava L.)Juice represents a source of antioxidant to inhibit lipid peroxidation and to improve erythrocyte membrane tenacity of Diabetes Mellitus treated rat. Needs continuation research to know mechanism of guava active agent job activity to other component in blood. Continuation research to guava effect to disease related to damage of cell membrane. And Continuation research about effective dose from kernel guava juice for Diabetes mellitus patient.
